Abstract
Objective: The aim of the present work was to formulate a tablet which disintegrates and dissolves rapidly to show a rapid onset of action. Orlistat used to manage obesity by reversibly inhibiting gastric and pancreatic lipases within the gut was selected as a model drug. Methods: In the present study, attempt has been made to prepare fast dissolving tablets of Orlistat using 3 different superdisintegrants like Sodium starch glycolate, croscarmellose sodium, and Plasdone by direct compression technique using 3 different concentrations of each superdisintegrant. The pre compression parameters of the prepared tablet blend like angle of repose, bulk density, tapped density, Carr’s index, Hausner’s ratio and the post compression parameters of the tablet like hardness, friability, weight variation, in vitro dispersion time, wetting time, water absorption ratio, in vitro disintegration time and in vitro drug dissolution were evaluated. Results: Angle of repose of powder blends for Orlistat was in the range of 29.82º±0.8 to 33.5±0.4, Carr’s index in between 10.90±0.7 to 26.21±0.8 and Hausner’s ratio less than 1.12±0.04. The above results indicate that the powder blends showed good flow properties. The overall results showed that the formulation is stable for a period at the above mentioned storage condition. It was further supported by FTIR analysis which showed that S9 had no interaction with excipients. S9 formulation was considered as the optimized formulation. Conclusion: It was concluded that formulation S9 showed better release characteristics of the drug. The stability studies were carried out for the optimized batch S9 for 30days and it showed acceptable results.
